Multi-substitutions of S. Singh's elucidated cocaine analog compounds

I've personally attempted contact with Mr. Singh as to what multiple-augmented variants have been attested to in the literature, their creation and any synthesis or activity facts discovered about them once made. I've tried twice now to contact him concerning literature beyond the cited paper used extensively on this page and whether he knows of any further publications in the academic field of the existence of such "muti-substitutions" (as Singh mentions, "combinatorial" approaches). The extreme of which, due to their efficacy may include the likes of the following when taken to the most possible alterations from the parent molecule:

Chemical derivative vs. structural analogue vs functional analog

I found this article confusing and realized that it appears to be conflating 3 different subjects:

1) Chemical derivatives of cocaine, see the Wikipedia article Derivative (chemistry)

2) Functional analogs of cocaine, see the article, Functional analog (chemistry)

3) Structural analogs of cocaine, see the article, Structural analog

Chemical derivatives can be naturally occurring or semi-synthetic. They may or may not have any overlapping biological effects similar to cocaine.

Functional analogs would include a vast array of naturally occurring, semi-synthetic, and synthetic compounds that would have some aspect of biologic effect as cocaine. It would include many substances that had no structural similarity to cocaine and aren't derived from cocaine. The list would be so long as to not be helpful.

Structural analogs would include synthetic compounds (such as fentanyl) or the Amino ester local anesthetics.

I think that the article would benefit from having these 3 meanings of "Cocaine analogue" clarified, but it would take multiple structural changes to the article. As a Wikipedia newby, I am reluctant to make this one of my first edits and I would appreciate comments/recommendations before doing any editing. Soloniuk (talk) 23:32, 28 October 2018 (UTC)[reply]

Functional analogs section - deserves reinstatement

There are referenced publishings, I note esp. the GBR compounds, that were created with the express purpose of being functional "cocaine analogs" and are referred to as such in the literature (see the references in the old reversions before May 25th '20). Perhaps at least a passing section for the historicity of what compounds have been created, functionally, as analogs to cocaine in particular and called by that name by their creators. _Nagelfar (talk) 16:18, 23 September 2020 (UTC)[reply]

overlay graphics with methylphenidate etc.

The former sections on methylphenidate included overlay pictures with cocaine analogs to show their similarity at binding. There is some such content lost in the sweeping change of taking out the functional section without saving those parts pertinent to cocaine and it's analogues QSAR specifically. _Nagelfar (talk) 16:25, 23 September 2020 (UTC)[reply]

Also, any link whatsoever with methylphenidate or the List of methylphenidate analogues has been omitted, yet we have mazidol derivates listed as functional/structural intermediates. This seems biased and I don't know the rationale of removing it (that it is a commonly prescribed medication and the stigma of cocaine as a relative functional adjunct?)
Esp. the part about: “the respective difference in their entropy of binding: cocaine being —5.6 kcal/mol & methylphenidate being —25.5 kcal/mol (Δs°, measured using [³H]GBR 1278 @ 25 °C)”[a] ← seems to be very pertinent to the QSAR of the relative substances and am a bit nonplussed to why these were removed.
